A Novel Selection System for Characterization and Identifcation of Arabidopsis Mutants Affected in Stress Responses

چکیده
In order to identify and characterize regulatory genes, able to control the expression of drought and salt responsive genes in Arabidopsis, a novel selection system has been designed. Several ten thousand transgenic Arabidopsis plants were created by efficient vacuum infiltration method that carries both the pRD29A-Luc or pADH1-Luc constructs and the pTAc17 tagging vector. T1 seedlings were screened for luciferase activity in the absence of stress. Our results suggests that the luciferase reporter system can be exploited for the identification and tagging of genes, the expression of which is positively or negatively regulated by ABA, salt or osmotic stress

In any organism that reproduces sexually, DNA Recombination plays vital roles in the generation of allelic diversity as well as in preservation of genome fidelity. Genome fidelity is particularly important in plants because mutations occurring during the development of flowering plants are heritable and can be passed onto the next generation. One of the gene families that play crucial roles in ...

To gain more insight into ABA signaling mechanisms, we conducted genetic screens searching for mutants with altered ABA response in germination and post-germination growth. We isolated seven putative ABA-hypersensitive Arabidopsis mutants and named them ABA-hypersensitive germination (ahg). These mutants exhibited diminished germination or growth ability on medium supplemented with ABA. We furt...

BACKGROUND Plants respond to many unfavorable environmental conditions via signaling mediated by altered levels of various reactive oxygen species (ROS). To gain additional insight into oxidative signaling responses, Arabidopsis mutants that exhibited tolerance to oxidative stress were isolated. We describe herein the isolation and characterization of one such mutant, oxt6. METHODOLOGY/PRINCI...

Phosphorus (P), in the form of phosphate ion (Pi), is a vital element contributing in biomolecule structures, metabolic reactions, signaling pathways and energy transfer within the living cells. The objective of the present study was to assess the influence of fungal infection on Pi metabolism in compare to the effects of phosphate stress in Arabidopsis. Quantification of total P contents showe...

Leaf senescence constitutes the last stage of leaf development in plants and proceeds through a highly regulated program in order to redistribution of micro- and macro-nutrients from the senescing leaves to the developing/growing plant organs. Initiation and progression of leaf senescence is accompanied by massive sequential alterations at various levels of leaf biology including leaf morpholog...
